DNA-binding, multivalent interactions and phase separation in transcriptional activation

چکیده
Transcription is an essential process in biology whereby gene-specific transcription factors target sites on DNA to recruit the basal machinery that will produce messenger RNA (mRNA). It a highly regulated multi-step involves many proteins and protein complexes. factors, mark genes for activation, other transcriptional regulators are enriched low-complexity disordered regions, which strongly linked multivalent binding phase separation. These regions can form dynamic complexes aspects of transcription. Many these separate vitro show evidence separation vivo. Whether interactions represent biologically relevant vivo controversial. However, what events do demonstrate co-cluster with vivo, forming clusters contribute events. We review some recently investigated consider how they our understanding

Multivalent Electrostatic Interactions and Autoinhibition Underlie Phase Separation of Npm1 with R-rich Nucleolar Protein Surf6
The nucleolus, the largest membrane-less organelle, is critical to cellular functioning as the site of ribosome biogenesis and as a fundamental cell stress sensor. It has previously been shown that the nucleolus assembles via liquid-liquid phase separation of its components from the surrounding nucleoplasm. Here, we show that two abundant nucleolar proteins often upregulated in certain cancers,...
متن کاملFrom DNA binding to transcriptional activation: Is the TALE complete?
How transcription factors (TFs) control enhancer and promoter functions to effect changes in gene expression is an important question. In this issue, Hau et al. (2017. J. Cell Biol. https://doi.org/10.1083/jcb.201701154) show that the TALE TF MEIS recruits the histone modifier PARP1/ARTD1 at promoters to decompact chromatin and activate transcription.
متن کاملA Phase Separation Model for Transcriptional Control
Phase-separated multi-molecular assemblies provide a general regulatory mechanism to compartmentalize biochemical reactions within cells. We propose that a phase separation model explains established and recently described features of transcriptional control. These features include the formation of super-enhancers, the sensitivity of super-enhancers to perturbation, the transcriptional bursting...
متن کاملControl of multivalent interactions by binding epitope density.
Receptor clustering by multivalent ligands can activate signaling pathways. In principle, multivalent ligand features can control clustering and the downstream signals that result, but the influence of ligand structure on these processes is incompletely understood. Using a series of synthetic polymers that vary systematically, we studied the influence of multivalent ligand binding epitope densi...
متن کاملMutations in the DNA-binding and transcriptional activation domains of v-Myb cooperate in transformation.
The v-Myb protein encoded by avian myeloblastosis virus causes oncogenic transformation of monoblastic cells committed to the monocyte/macrophage lineage. v-Myb is a doubly truncated form of its normal cellular counterpart, c-Myb. In addition to its N- and C-terminal deletions, v-Myb contains a number of amino acid substitutions relative to c-Myb. We have previously shown that neither overexpre...
